本文介绍了使用单一混合试剂的终点法和LAL显色动力学法。该试剂可用缓冲液、鲎试剂和底物(显色鲎试剂)混合冻干或用市售的LAL凝胶试剂、底物和缓冲液(显色法、凝胶法通用)混合后立即使用。这种单一试剂既可用于显色动力学方法,也可用于一步终点法(规定孵育时间)。由于去除了许多步骤和技术误差,因此可极大地减少显色法的差异。本文对动力学法和终点法进行了详细地比较,单一试剂法也与传统的多试剂法进行了对比。
This article describes the endpoint method and the LAL chromogenic kinetic assay using a single mixed reagent. The reagent can be mixed with buffer, reagent and substrate (chromogen reagent), freeze-dried or mixed with commercially available LAL gel reagent, substrate and buffer (chromogenic method, gel method) immediately. This single reagent can be used both for chromogenic kinetics and one-step end-labeling (for incubation times). By removing many of the steps and technical errors, the differences in colorimetric methods can be greatly reduced. In this paper, dynamical method and end-point method are compared in detail. The single reagent method is also compared with the traditional multi-reagent method.
