目的 研究人垂体腺瘤中雌激素受体基因 (estrogenreceptorgene ,ER)mRNA与蛋白水平的表达及其与垂体腺瘤生物学特性的关系。方法 用免疫组化法和逆转录 聚合酶链反应 (RT PCR法 ) ,检测 6 0例各种类型垂体腺瘤标本中ER蛋白的表达 ,比较不同类型垂体腺瘤ER蛋白表达的差异及ER蛋白表达与mRNA表达的差异。结果 泌乳素腺瘤、促性腺激素细胞瘤中均有ER蛋白表达 ,其表达与mRNA表达一致 ;无功能腺瘤中 2例有ER蛋白表达 ,其蛋白表达与mRNA表达不一致 ;其它类型垂体腺瘤未发现ER基因表达。结论 ER基因表达的检测可有助于评价PRL腺瘤的增殖活性及筛选抗雌激素治疗对象。ER基因的表达异常可能与PRL腺瘤及促性腺激素细胞瘤的发生、发展有关
Objective To study the expression of estrogen receptor gene (ER) mRNA and protein in human pituitary adenomas and its relationship with biological characteristics of pituitary adenomas. Methods The expression of ER protein in 60 specimens of pituitary adenomas was detected by immunohistochemistry and reverse transcription polymerase chain reaction (RT PCR). The differences in ER protein expression and ER protein expression in different types of pituitary adenomas were compared. Expression and mRNA expression differences. Results The expression of ER protein in prolactin adenoma and gonadotropin cell tumor was consistent with the expression of ER protein. The expression of ER protein was found in 2 cases of nonfunctioning adenoma. The expression of ER protein was inconsistent with that of mRNA expression; other types of pituitary adenoma No ER gene expression was found. Conclusion The detection of ER gene expression may be helpful in evaluating the proliferative activity of PRL adenomas and screening anti-estrogen therapeutic targets. Abnormal expression of ER gene may be related to the occurrence and development of PRL adenoma and gonadotropinoma
