利用人脐带血单个核细胞,联合粒/巨噬细胞集落刺激因子(GM-CSF)和肿瘤坏死因子-α(TNF-α),经2周左右可培养出大量树突状细胞(DCs);来自正常人外周的单个核细胞,经GM-CSF和IL-4作用,8-10天后亦可培养出大量(4-5)×10~6/50ml)DCs。该研究为进一步研究DCs负载肿瘤抗原作为疫苗,为肿瘤免疫中的应用奠定了物质基础。
Using mononuclear cells of human umbilical cord blood, GM-CSF and tumor necrosis factor-α (TNF-α), a large number of dendritic cells (DCs) can be cultured after about 2 weeks; Mononuclear cells from normal human peripheral blood can be cultured in a large amount (4-5) × 10 ~ 6 / 50ml) after 8-10 days by GM-CSF and IL-4. This study lays the material foundation for the application of tumor immunity in order to further study the DCs loaded with tumor antigens as a vaccine.
