目的研究大黄、虫对大鼠子宫肌瘤模型孕激素及其受体的影响。方法将70只SD大鼠随机分为7组:正常对照组、模型组、大黄虫小剂量组、大黄虫中剂量组、大黄虫大剂量组、大黄虫丸组、桂枝茯苓胶囊组,采用雌孕激素负荷法建立大鼠子宫肌瘤模型,放射免疫法测定大鼠血清孕激素(P)水平,免疫组织化学染色观察大鼠子宫平滑肌细胞孕激素受体(PR)的表达。结果模型大鼠血清中P水平,子宫平滑肌细胞PR的表达明显高于正常大鼠,大黄虫可降低血清中P水平,抑制平滑肌细胞PR的表达。结论P及PR在子宫肌瘤生长中起重要作用,这可能是大黄虫治疗子宫肌瘤的机理之一。
Objective To study the effects of rhubarb and pinworm on progesterone and its receptor in rat uterine fibroids model. Methods Seventy SD rats were randomly divided into 7 groups: normal control group, model group, rhubarb small dose group, rhubarb worm medium dose group, rhubarb worm large dose group, rhubarb worm group, Guizhi Fuling Capsule group, the rat uterine fibroids model was established by estrogen and progesterone loading, the level of serum progesterone (P) was determined by radioimmunoassay, and the expression of progesterone receptor (PR) in rat uterine smooth muscle cells was observed by immunohistochemical staining . Results The level of P in serum of rat model rats and the expression of PR in uterine smooth muscle cells in model rats were significantly higher than those in normal rats. Rhubarb could reduce the level of P and inhibit the expression of PR in smooth muscle cells. Conclusion P and PR play an important role in the growth of uterine fibroids, which may be one of the mechanisms of Rhubarb in the treatment of uterine fibroids.
