目的 :观察风疹病毒 ( RV)分离株在人的神经细胞中形态和形态发生过程。方法 :利用超薄切片电子显微镜技术对自行分离的 RV JR2 3 株在人脑神经细胞中的形态与形态发生过程进行了研究。 结果 :RV JR2 3 株感染人脑神经细胞 2 4 h后 ,可在细胞浆的核周围观察到病毒相关结构 ,包括电子致密颗粒 ,1 0 nm,核衣壳样结构 ,1 5~ 2 5nm,“包涵体”样结构 ,“条纹”样结构伴随核衣壳样颗粒 ,并逐渐增多。 96h可见完整病毒颗粒 ,1 92 h达到高峰。病毒颗粒呈圆形 ,由单层脂质膜包绕 ,直径 60~ 80 nm。病毒在双层核膜中间装配 ,然后披上单层核膜而成熟 ,核膜破碎后释放入细胞浆。病毒颗粒多见于核周围。结论 :RV JR2 3 株能在人神经细胞中广泛增殖 ,其形态发生有独特的方式。
Objective: To observe the morphology and morphogenesis of RV isolates in human neural cells. Methods: Morphology and morphogenesis of RV JR2 3 strain isolated from human brain were studied by ultrathin section electron microscopy. Results: The virus-associated structure was observed around the nucleus of cytoplasm after infection of human brain neurons with RV JR2 3 for 24 h, including electron-dense particles, 10 nm nucleocapsid-like structures, 15-25 nm, “Inclusion body” -like structure, “stripe” -like structure with nucleocapsid-like particles, and gradually increased. 96h visible complete virus particles, peaked at 1 92 h. The virus particles are round, surrounded by a single lipid membrane diameter of 60 ~ 80 nm. Virus in the middle of the double nuclear membrane assembly, and then put on a single nuclear membrane mature and nuclear membrane broken into the cytoplasm. Virus particles more common in the nucleus. Conclusion: RV JR2 3 strain can widely proliferate in human nerve cells, and its morphogenesis has a unique way.
