不同冠状病毒非结构蛋白16发挥2-0甲基转移酶功能的机制研究

  高等真核生物细胞信使RNA (mRNA)及一些病毒RNA的5-端均具有N7或者2-O甲基化修饰的帽子结构。这种帽子修饰具有很多重要的生物学功能:保证信使RNA的稳定性、剪切、核输出及有效的翻译等。除此以外,近期国际上发现冠状病毒nsp16编码的2-O甲基转移酶对病毒5帽子结构进行的2-O甲基化修饰,可以使病毒核酸逃避胞内MDA5受体(识别病毒双链RNA)的免疫识别或者逃避干扰素诱导蛋白(IFIT)介导的免疫抑制。又由于病毒本身的加帽与细胞内信使RNA的加帽过程及机制存在很大差别,使得病毒加帽酶作为抗病毒药物靶标成为可能。近期暴发的与SARS类似的新型冠状病毒——中东呼吸综合征冠状病毒(MERS-CoV),高致死率使得冠状病毒的研究更有意义。
