Objective To study the anti-proliferative mechanism of liver X receptors (LXRs) agonist TO901317 on human breast cancer cells, so as to provide a new basis for the diagnosis and treatment of human breast cancer.Methods Immunostaining analysis were used to detect the expression of LXRα, NF-κB p65 and CyclinD1 in human breast cancer and paired normal adjacent human breast tissues.MCF-7 human breast cancer cells were treated with different concentration of TO901317 (0,5, 10, 20μM) for different time (0, 6, 12, 24, 48h).MTT and flow cytometry assay were used to detect the proliferation of MCF-7 cells.The expression of LXRα, NF-κB p65,cyclin D 1 was examined by reverse transcription-polymerase chain reaction (RT-PCR) and western blot.Then, the effect of LXRα siRNA and Pyrrolidinedithiocarbamic acid (PDTC, an inhibitory of NF-κB) on TO901317 were observed respectively.
