Due to low expression of coxsackie-adenovirus recptor (CAR) in leukemia cells, the utility of type 5 adenoviruses (Ad5) in leukemic treatment is limited.We constructed recombinant sCAR-ligand fusion proteins including sCAR-IL3 and sCAR-4NI, which successfully transduced Ad5 into CD123+ or CD47+ leukemia cells.Based on this observation, oncolytic adenoviruses Ad.sp-EIAsCAR-IL3 and Ad.sp-E1A-sCAR-4NI were constructed, in which the viral E1A was controlled by a survivin promoter, and sCAR-IL3 or sCAR-4N1 fusion gene expression cassette was inserted into the viral genome.
