Hematopoietic stem cell (HSC) transplantation is becoming an important therapeutic strategy for the treatment of cancer, congenital immunodeficiencies, inborn errors of metabolism, and various hematological diseases.The widespread use of various treatment protocols utilizing HSCs have resulted in an increased demand for cryobiological techniques for storing these cells.This need has led to a reexamination of many cryobiological practices.So far, HSCs are cryopreserved with a robust system named cooling rate controlled freezer(CryoMedTM).This device can cool the samples with a specific cooling rate or execute some complex cooling profiles by adjusting the flux of liquid nitrogen (LN2), and record the temperature history of the samples.However, this device is very expensive and the LN2 consumption is very high.Some other simple protocols have also been used for cell suspensions, such as the samples being placed in Styrofoam boxes and placed into-40℃ freezers, vapor nitrogen environments, or a container with alcohol baths and then in a-80℃ freezer.These systems are cheap and with low or without LN2 consumption.
