Since the discovery of dopamine as a chemical neurotransmitter in the brain, many human neurological disorders have been linked to the malfunction of this important neural system. For example, Parkinsons disease is caused by the selective degeneration of dopamine neurons in the substantia nigra, and dopamine system is strongly implicated in the development of drug and alcohol addiction. The mechanisms underlying dopamine neuron development, regeneration, degeneration, and function are not well understood, and as a consequence, effective treatments for the related human disorders are largely lacking. In this talk, we will discuss our use of a simple vertebrate model system, the zebrafish Danio rerio, to study mechanisms underlying dopamine neuron development, degeneration, and to develop cellular and behavioral assays for functional gene discovery and small molecule screening.
