This study was designed to investigate the effect of the BAX inhibitor-1 (BI-1)/testis-enhanced gene transcript (TEGT) on the regulation of cellular Ca2+ and its related effect on apoptosis.BI-1 acts as a Ca2+ channel on endoplasmic reticulum (ER) membrane with a putative pH-sensing motif,EKDKKKEKK.Under acidic pH conditions,ER and reconstituted lipid membrane assays revealed that BI-1 released tremendous amounts of Ca2+,making BI-1-overexpressing cells more sensitive to acidic pH-induced mitochondrial Ca2+ and BAX accumulation and cell death.In a normal pH range,BI-1 increases Ca2+ permeability from the ER,regulating mitochondrial BAX oligomerization,enhancing the threshold of mitochondrial membrane transition pore opening and leading to cell-protective effects.
