Tumor necrosis factor Related Apoptosis Inducing Ligand (TRAIL) has been shown to trigger apoptosis in a variety of human tumor cells by signaling through its two physiological cell surface receptors,TRAIL-R1 (DR4) and TRAIL-R2 (DR5).The heightened interest in the clinical use of TRAIL stems from the restricted expression of DR4 and/or DR5 on tumor cells,but not normal cells.However,development of resistance to TRAIL remains a serious problem in TRAIL responsive tumors,and involves mechanisms that include receptor internalization or downregulation and/or inhibition of death signaling at post-mitochondrial level.
