The experiment was conducted to explore the effects of feeding Saanen goat E.adenophorum-containing feedstuffs on apoptosis, cell cycle progression, and relative expression quantity of apoptosis genes of Bax, Bcl-2,caspase-3, 8, 9 in kidney through flow cytometry and qRT-PCR.Sixteen Saanen goats randomly divided into four groups were fed on control diet (non-E.adenophorum) and experimental diets mixed with 40%, 60%, 80% E.adenophorum leaves for 3 months respectively.As tested by flow cytometry, the results showed that the percentage of apoptosis in renal cell was increased and the normal renal cells obviously were decreased in experimental groups as compared to control.Renal cells in G0/G1 phase were much higher, and lower in S phase and proliferation index value in experimental groups which means E.adenophorum importantly arrest the cell cycle transition from G0/G1 to S phase which made renal cell cant complete the cell cycle activity and increased cellular apoptosis in the kidney.qRT-PCR results suggested that E.adenophorum inhibited Bcl-2 expression, promoted Bax translocation and caspase-3, 9 activation in renal cells which means some pro-apoptotic genes expressed and apoptosis suppressor gene was inhibited, However, caspase-8 activity did not exhibit significant changes.The findings indicate that E.adenophorum probably has dose-dependent nephrotoxicity effect on goat as it markedly induced apoptosis.
