壬基酚干扰17β-雌二醇对豚鼠缺血再灌注损伤心肌保护作用的研究

  目的 壬基酚(nonylphenol,NP)是环境内分泌干扰物的一种,因具有强大脂溶性而被广泛应用于工业、纺织业及生活用品中,目前对环境的污染甚为严重。众多研究证实NP长期积蓄于生物体内能对生殖系统、内分泌系统及心血管系统等产生不可忽视的毒性作用,而对其急性毒性作用的研究尚不明确。方法 本研究首先将10-7 M NP和10-5 M NP直接灌流离体心脏,观察10-7 M NP和10-5 M NP对离体心脏心率(heart rate, HR)、冠脉流量(coronary flow,CF)、乳酸脱氢酶(lactate dehydrogenase,LDH)的影响;其次将10-7 M NP和10-5 M NP作用于离体心脏缺血再灌注模型中观察HR、CF、LDH、肌酸激酶(creatine kinase,CK)、梗死面积和超氧化物歧化酶(superoxide dismutase,SOD)等指标,探讨NP是否能够加重缺血再灌注损伤;最后将10-7 M NP和10-5 M NP分别与10-7 M 17β-雌二醇(17β-estradiol,E2)联合作用于缺血再灌注模型观察HR、CF、LDH、CK、梗死面积和SOD等指标,探讨NP是否能够干扰E2对缺血再灌注损伤心肌保护作用。
