Octodonta nipae(Maulik)(Coleoptera: Chrysomelidae)is a serious invasive insect pest of palm plants in southern China.Tetrastichus brontispae Ferrière(Hymenoptera: Eulophidae)is a natural enemy of this pest and can serve as an important biological control agent.Encapsulation is an important cellular response in insect innate immunity, and it is triggered by the eggs of parasitic wasps. In order to reveal the molecular mechanism of encapsulation process of O.nipae parasitized by T.brontispae, the cDNAs of beta-integrin,calreticulin and Rac1 from O.nipae were isolated and their transcription altered by parasitization of T.brontispae was investigated. The beta-integrin,calreticulin and Rac1 genes were 2056bp,1538bp and 3467 bp long, with open reading frames of 1797bp,1212bp and 579 bp,encoding 598,403 and 192 amino acids, respectively.Quantitative real time PCR analyses indicated that the transcript levels of beta-integrin,calreticulin and Rac1 in O.nipae pupae were significantly suppressed after parasitization by T.brontispae.These results suggest that the down-regulation of these encapsulation-related genes of O.nipae may be related to the immune defense strategy of T.brontispae protecting its offspring against host encapsulation.
