乙酰胆碱与去甲肾上腺素对食管癌细胞分化的调制作用及其机制

  研究背景 食管癌的病因与发病机制尚不清楚,食管癌变具有双向发展的不稳定性和受神经系统功能状态的调节.目的探讨食管神经和递质对食管上皮细胞癌变的作用与机制,为防治食管癌变与复发提供实验依据.方法:采用50具尸体标本食管神经解剖与食管癌细胞系体外培养,将食管癌细胞系EC109细胞分为乙酰胆碱(ACh)组、ACh加阿托品组、去甲肾上腺素(NE)组、NE加托拉唑林组、ACh+NE组、对照组,体外培养14d,经HE和氧化还原修复酶-1(PRX1)免疫组化染色、MTT、细胞平板克隆、流式细胞术等技术方法,并对结果进行统计学处理.结果 (1)食管胸部中下段迷走神经分布密度较低;(2)各药物处理组与对照组相比,①细胞3d长出突起,7d明显长长,14d部分细胞之间相互连结形成网状;②PRX1免疫组化染色明显加深;③核质比下降;④细胞增殖活性下降,细胞增长抑制率提高,细胞多数阻滞于细胞周期的G1期,凋亡细胞数目增多.各组的指标与对照组者相比均有显著性差异(P<0.05).结论 ①神经递质ACh和NE对食管癌细胞分化具有诱导作用,其作用可能与其增强DNA修复酶的表达有关;②ACh和NE对食管癌细胞增殖具有抑制、对凋亡具有促进作用;③结合食管胸部中下段食管神经分布密度较低,可能存在生理性神经功能薄弱区,而成为食管癌变易感的神经性因素之一.该发现为开拓食管癌发病机制和防治研究的新领域提供了实验依据.
