人VEGF受体II基因3区的克隆、表达研究

用RT PCR法从人胎儿脐静脉内皮细胞中克隆人VEGF受体II基因 3区 ,将其重组到 pAYZ表达载体中 ,构建成人VEGF受体II基因 3区表达载体。将该载体转化大肠杆菌 16C9,获得稳定表达 ,表达产物以可溶性状态存在 ;SDS PAGE和Westernblot分析显示 ,其分子量约为 14kD。以纯化的表达产物免疫Balb/c小鼠 ,眼球采血制备抗血清 ,抗血清效价在 10 3以上 ,并具有抑制VEGF诱导内皮细胞增殖的作用 ,在抑制肿瘤血管新生中具有潜在的应用前景。 The third region of human VEGF receptor II gene was cloned from human embryonic umbilical vein endothelial cells by RT-PCR and recombined into pAYZ expression vector to construct the expression vector of human VEGF receptor II gene region 3. The vector was transformed into Escherichia coli 16C9 to obtain a stable expression, the expression product exists in a soluble state; SDS PAGE and Western blot analysis showed that the molecular weight of about 14kD. Balb / c mice were immunized with the purified expression product. Blood was collected from the eyeball to prepare antiserum. The titer of the antiserum was more than 10 3. The antisera could inhibit the proliferation of endothelial cells induced by VEGF and have potential application in the inhibition of tumor angiogenesis.
