通过研究1型糖尿病(T1DM)大鼠肾皮质和正常大鼠肾皮质蛋白质表达的差异,进一步探讨T1DM引起肾脏病变的可能机制。实验采用链脲佐菌素(STZ)诱发SD大鼠产生高血糖,建立T1DM模型,应用双向凝胶电泳技术分离大鼠肾皮质蛋白。电泳图谱结果:正常组分辨出蛋白斑点468±34个;DM组分辨出蛋白斑点423±23个,其中差异显著的蛋白斑点43个,与正常组比较,DM组有15个蛋白斑点表达上调,24个表达下调,2个失表达,2个蛋白斑点只在DM组表达。本实验证明了T1DM大鼠肾皮质蛋白质表达发生了显著变化,差异蛋白质点的进一步鉴别研究将有助于探讨T1DM肾病的发病机制。
To explore the possible mechanism of T1DM-induced nephropathy by studying the differences of protein expression in renal cortex of type 1 diabetic rats (T1DM) and normal rats. Experimental use of streptozotocin (STZ) induced SD rats produce high blood sugar, the establishment of T1DM model, the use of two-dimensional gel electrophoresis separation of rat renal cortex protein. The result of electrophoresis showed that 468 ± 34 protein spots were identified in normal components, 423 ± 23 protein spots were found in DM component, and 43 protein spots were significantly different among them. Compared with normal group, 15 protein spots were up-regulated in DM group, 24 downregulation, 2 loss expression, 2 protein spots were only expressed in DM group. This experiment demonstrated that there was a significant change in renal cortical protein expression in T1DM rats. Further identification of differential protein spots will be helpful to explore the pathogenesis of T1DM nephropathy.
