本文报道了一种从感染曼氏血吸虫或日本血吸虫的小鼠肠中分离大量无菌血吸虫虫卵的方法。将感染波多黎各株曼氏血吸虫或台湾株日本血吸虫的小鼠在感染后7~8周注射0.2ml硫喷妥钠致死。取肠用自来水冲洗。割弃大肠,纵向切开小肠,刮去肠内容物和粘膜层,并除去肠系膜组织及周围的脂肪组织。将小肠浸泡在0.2M的氯化钠溶液中,在室温下过夜。第2天,以10条肠子为一批悬浮于100ml的0.2M盐水中,在华林氏搅拌器(Model 31BL46)中连续迅速地粉碎2次,每次20秒钟、两次间隔10秒钟。将匀浆立即倒入直径为20cm的40号美国标准筛子(筛孔为420mμ)上,用500ml盐水冲洗。将全部过筛后的小肠匀浆再依次用:40号(420mμ)、80号(177mμ)、100号(149mμ)、200号(74mμ)和325号(44mμ)筛子进行过筛。这些筛子约用1L的盐水冲洗。虫卵能通过40~200号筛子而不能通过325号筛子,在325号筛子中收集虫卵,再将这些虫卵用500ml盐水悬浮后,用上述的一堆筛子进行重复过筛。将从325号筛子上收集的虫卵用无菌
Here we report a method for isolating a large number of sterile Schistosome eggs from mouse gut infected with Schistosoma mansoni or Schistosoma japonicum. Mice infected with S. pombe or S. japonicum were injected with 0.2 ml of thiopental 7 to 8 weeks after infection. Take intestinal wash with tap water. Discard the large intestine, incise the small intestine longitudinally, scrape off the intestinal contents and mucosa, and remove the mesentery tissue and surrounding adipose tissue. The small intestine was soaked in 0.2 M sodium chloride solution at room temperature overnight. On the second day, 10 intestines were suspended in 100 ml of 0.2 M saline in a batch and continuously and rapidly crushed twice in a Waring blender (Model 31BL46) twice for 20 seconds at intervals of 10 seconds . The homogenate was immediately poured onto a No. 40 U.S. standard sieve (mesh size 420 mμ) with a diameter of 20 cm and rinsed with 500 mL of saline.
