目的 研究胰岛素样生长因子I受体 (IGF IR)基因表达对脐血T淋巴细胞功能的影响及其临床意义。方法 利用脂质体将IGF IR反义寡核苷酸 (ASON)转入脐血淋巴细胞 ,通过同位素掺入法了解淋巴细胞增殖功能 ,采用逆转录聚合酶链反应技术检测IGF IR、IL 2、IL 4和干扰素 (IFN)r的mRNA表达水平。结果 通过IGF IRASON可抑制脐血淋巴细胞IGF IRmRNA的表达约 5 0 % ,同时淋巴细胞的增殖功能较单纯PHA刺激也下降 5 0 %左右 ;细胞因子IL 2、IL 4、IFNr的mRNA水平也有不同程度的降低 ,其中IL 2和IFNr的mRNA水平经IGF IRASON作用后较单纯PHA刺激相比降低幅度约为 30 %~ 5 0 % ,IL 4mRNA降低幅度约为 2 0 %~ 30 %。结论 IGF IR参与脐血淋巴细胞的增殖和细胞因子的产生 ,提示在维持细胞免疫功能和调节婴儿时期TH1 TH2应答相对平衡的过程中IGF IR可能具有重要意义。
Objective To study the effect of insulin-like growth factor-I receptor (IGF IR) gene expression on the function of cord blood T lymphocytes and its clinical significance. Methods IGF IR antisense oligonucleotide (ASON) was transfected into umbilical cord blood lymphocytes by liposome, lymphocyte proliferation was detected by isotope incorporation method, and the expression of IGF IR, IL 2, IL 4 and interferon (IFN) r mRNA expression levels. Results IGF IRASON inhibited the expression of IGF IR mRNA in cord blood lymphocytes by about 50%, while the proliferation of lymphocytes decreased by 50% compared with that of PHA alone. The mRNA levels of cytokines IL 2, IL 4 and IFNr were also different The level of IL-2 mRNA and IFN-γ mRNA decreased by 30% ~ 50% compared with that of pure PHA stimulated by IGF IRASON, while the decrease of IL 4 mRNA was about 20% ~ 30%. Conclusion IGF IR is involved in the proliferation of cord blood lymphocytes and the production of cytokines, suggesting that IGF IR may play an important role in maintaining cellular immune function and regulating the relative balance of TH1 TH2 response in infancy.
