为研究 bFGF(碱性成纤维细胞生长因子)对辐射诱导的血管内皮细胞凋亡的作用及其信号转导途径,以 Coγ 射线照射法建立人脐静脉内皮细胞株 ECV304 细胞凋亡模型,以流式细胞术(Annexin-V-FITC+PI 标 60记)检测细胞凋亡率。观察不同浓度的 bFGF 和 MEK 的特异性抑制剂 U0126 对细胞凋亡的影响。结果显示,bFGF 对辐射诱导的 ECV304 细胞凋亡有明显的抑制作用,而 U0126 可阻断 bFGF 的抗凋亡作用。提示 bFGF可能通过 MAPK 信号转导途径抑制辐射诱导的血管内皮细胞凋亡。
To investigate the effect of bFGF (basic fibroblast growth factor) on radiation-induced apoptosis of vascular endothelial cells and its signal transduction pathway, the apoptosis model of human umbilical vein endothelial cell line ECV304 was established by Co γ ray irradiation. Cell apoptosis was detected by Annexin V-FITC + PI-60 assay. The effects of different concentrations of bFGF and MEK specific inhibitor U0126 on apoptosis were observed. The results showed that bFGF significantly inhibited the apoptosis of ECV304 cells induced by radiation, while U0126 blocked the anti-apoptotic effect of bFGF. These results suggest that bFGF may inhibit radiation-induced apoptosis of vascular endothelial cells through the MAPK signal transduction pathway.
