[目的]比较分析人大肠癌细胞HSP70活化CIK细胞和普通CIK细胞的杀瘤活性,为该类CIK的临床应用提供有重要参考价值的实验依据。[方法]用液相色谱法提纯人大肠癌细胞中的HSP70。取健康人外周血单核细胞,用CD3单克隆抗体、IFN-γ、IL-1α、IL-2等细胞因子对其诱导、培养获取CIK细胞;运用GM-CSF、IL-4、TNF-α等细胞因子对其诱导、培养获取DCs,HSP70诱导活化DCs,按HSP70活化的DC:CIK=1:3混合培养1周。用MTT法进行杀瘤活性试验。[结果]人大肠癌细胞HSP70活化的CIK细胞和普通CIK细胞的杀瘤活性相比较,在效靶比例为10:1、20:1、40:1时,杀伤活性差异均具有统计学意义(P<0.01)。[结论]与普通的CIK细胞相比,肿瘤细胞HSP70活化的CIK细胞具有更强的杀瘤活性。
[Objective] To comparatively analyze the tumoricidal activity of HSP70-activated CIK cells and ordinary CIK cells in human colorectal cancer cells, and provide an experimental basis for the clinical application of such CIKs with important reference value. [Method] The purification of HSP70 in human colorectal cancer cell by liquid chromatography. Peripheral blood mononuclear cells (PBMCs) were obtained from healthy volunteers. CIK cells were induced by culturing CD3 monoclonal antibody, IFN-γ, IL-1α and IL-2 cytokines. GM-CSF, IL-4 and TNF- DCs were induced by culturing and culturing. DCs stimulated by HSP70 and DCs CIK = 1: 3 activated by HSP70 were mixed and cultured for 1 week. The killing activity test was carried out by MTT method. [Result] The killing activity of HSP70-activated CIK cells in human colorectal cancer cells was significantly lower than that of normal CIK cells (10: 1, 20: 1, 40: 1) P <0.01). [Conclusion] Compared with normal CIK cells, CIK cells activated by HSP70 in tumor cells have stronger antitumor activity.
