给BAlB/C小鼠注射HBsAg,取脾细胞与Sp2/O瘤株融合,经ELISA和PHA方法检测杂交瘤上清获得19个阳性孔。经4次克隆化,建立了2株分泌抗-HBs的杂交瘤。2个月在体外传代6次,液氮中经4次反复冻融仍稳定地分泌抗体。接种BALB/C小鼠腹腔后产生高滴度抗体腹水。 1975年kohler和Milstin首创杂交瘤技术以来,Shih,Wands和郭恒昌先后在国外和国内报道建立分沁抗-HBs的细胞系。我们实验室应用这一技术将亲和层析纯化得到的HBsAg免疫小鼠亦获得2株抗-HBs单克隆抗体,现报告如下;
BAlB / C mice were injected with HBsAg, spleen cells were fused with Sp2 / O tumor strains, and 19 positive wells were detected by ELISA and PHA. After four clones, two anti-HBs secreting hybridomas were established. 2 passages in vitro 6 times, liquid nitrogen in 4 repeated freezing and thawing still stable secretion of antibodies. BALB / C mice were inoculated intraperitoneally with high titers of antibody ascites. Shih, Wands and Guo Hengchang reported on the establishment of the cell line of anti-HBs in foreign and domestic since 1975 when Kohl and Milstin pioneered the hybridoma technology. Our laboratory using this technology will be purified by affinity chromatography and HBsAg-immunized mice also obtained 2 anti-HBs monoclonal antibody, are reported as follows;
