【目的】研究半枝莲多糖(Scutellaria barbata polysaccharides,SBP)对肝癌细胞SMMC7721生长的抑制及相关机制。【方法】将不同浓度SBP作用于SMMC7721肝癌细胞,采用四甲基偶氮唑盐(MTT)法检测半枝莲多糖对肝癌细胞SMMC7721生长增殖的抑制作用;实时荧光定量聚合酶链反应(Real-time PCR)检测细胞周期蛋白依赖性激酶(CDK2)与转录因子E2F1 mRNA表达;蛋白印迹法(Western blot)检测CDK2与E2F1蛋白表达。【结果】SBP对肝癌细胞SMMC7721具有明显的生长抑制作用,其中10、2.5μg/mL SBP组作用显著,均可使CDK2与E2F1 mRNA和蛋白表达下调。【结论】SBP对肝癌细胞SMMC7721具有增殖抑制作用,其机制可能与CDK2和E2F1表达下降有关。
【Objective】 To investigate the inhibitory effect of Scutellaria barbata polysaccharides (SBP) on the growth of hepatocellular carcinoma cell line SMMC7721 and its related mechanisms. 【Method】 SMMC7721 hepatoma cells were treated with different concentrations of SBP. The inhibitory effect of Scutellaria barbata polysaccharide on the growth and proliferation of hepatocellular carcinoma SMMC7721 cells was detected by MTT assay. Real-time fluorescent quantitative polymerase chain reaction (Real- time PCR was used to detect the expression of cyclin-dependent kinase (CDK2) and transcription factor E2F1 mRNA. The expressions of CDK2 and E2F1 protein were detected by Western blot. [Results] SBP had obvious growth inhibitory effect on SMMC7721 hepatocellular carcinoma cells, of which 10 and 2.5μg / mL SBP group had significant effect, both of which could down-regulate the expression of CDK2 and E2F1 mRNA and protein. 【Conclusion】 SBP can inhibit the proliferation of SMMC7721 hepatocellular carcinoma cells, and its mechanism may be related to the decrease of CDK2 and E2F1 expression.
