目的:研究肌动蛋白相互作用蛋白1(AIP1)在人肝细胞癌(HCC)中的表达情况,并探究其表达与上皮间质转化(EMT)的相关性。方法:选取6个肝癌细胞系和正常的肝细胞系L02,以及40对HCC组织和配对的癌旁非瘤肝组织标本,通过qRT-PCR检测AIP1 mRNA的表达,western blot检测AIP1蛋白的表达,最后采用免疫组织化学染色(IHC)验证组织中AIP1的表达并分析其表达与EMT标志物的关系。结果:肝癌细胞系中AIP1的mRNA和蛋白表达水平都显著高于正常肝细胞系L02。同样在新鲜冰冻的组织标本中,肝癌组织中AIP1的m RNA和蛋白表达都显著高于相应的癌旁非瘤肝组织,且通过IHC得到了进一步的证实。最后,IHC结果发现AIP1的上调可能与肝癌的EMT现象相关。结论:AIP1在肝癌中的表达发生上调,并可能通过促进肝癌的EMT而促进肝癌的侵袭转移。
AIM: To investigate the expression of actin-interacting protein 1 (AIP1) in human hepatocellular carcinoma (HCC) and to investigate its relationship with epithelial-mesenchymal transition (EMT). Methods: Six liver cancer cell lines and normal liver cell line L02, as well as 40 pairs of HCC tissues and matched paracancer non-tumor liver tissues were selected. The expression of AIP1 mRNA was detected by qRT-PCR and the expression of AIP1 protein was detected by western blot. Finally, immunohistochemical staining (IHC) was used to verify the expression of AIP1 in the tissue and to analyze the relationship between its expression and EMT markers. Results: The mRNA and protein expression of AIP1 in hepatocellular carcinoma cell lines were significantly higher than that in normal liver cell line L02. Similarly in fresh frozen tissue samples, the m RNA and protein expression of AIP1 in HCC tissues were significantly higher than that of the corresponding non-cancerous non-tumorous liver tissues and was further confirmed by IHC. Finally, IHC results showed that AIP1 upregulation may be related to the EMT phenomenon of liver cancer. Conclusion: The expression of AIP1 is up-regulated in hepatocellular carcinoma and may promote the invasion and metastasis of hepatocarcinoma by promoting the EMT of hepatocellular carcinoma.
