将用CPV免疫的BALB/c小鼠脾细胞与SPV2/0细胞在聚乙二醇作用下融合,用HI法筛选,有限稀释法克隆三次,得到6株分泌抗CPVMcAb的杂交瘤细胞株,其中4株分泌IgG1,2株分泌IgM。将杂交瘤细胞注射BALB/c小鼠诱生腹水,摸索出了大量生产腹水的最佳实验条件。采用CPV McAb研制触CPV快速诊断试剂盒,检测47份病犬粪样,在30min即可肉眼判定结果,与HA法和常规ELISA相比,具有简便、快速、特异之优点。应用CPVMcAb治疗CPV发病犬287只,治愈率高达97.21%,显著高于其他疗法的治疗效果,治疗机理除对体内病毒的中和作用外,有可能促进机体的主动免疫反应。在对46只幼犬进行CPV疫苗免疫时,使用CPVMcAb的23只,其免疫成功率为100%,而未使用CPVMcAb的23只对照犬,其免疫成功率仅为78.3%,表明CPV McAb在疫苗免疫中可起积极作用。
The BALB / c mouse spleen cells immunized with CPV were fused with SPV2 / 0 cells under the action of polyethylene glycol, screened by HI method and cloned by limiting dilution three times to obtain six hybridoma cell strains secreting anti-CPVMcAb 4 strains secreted IgG1 and 2 strains secreted IgM. BALB / c mice were injected with hybridoma cells to induce ascites, and the optimal experimental conditions for mass production of ascites were explored. CPV McAb was used to develop a rapid CPV diagnostic kit. 47 cases of canine dung samples were detected, and the result of naked eye examination was determined within 30 minutes. Compared with HA and conventional ELISA, the method was simple, rapid and specific. The application of CPVMcAb in the treatment of CPV dogs 287, the cure rate as high as 97.21%, significantly higher than the therapeutic effect of other therapies, in addition to the mechanism of treatment of the virus in vivo neutralization, it may promote the body’s active immune response. Twenty-three puppies CPV vaccine immunized with CPVMcAb had an immunization success rate of 100%, whereas 23 control dogs without CPVMcAb had a vaccine success rate of only 78.3%, indicating that CPV McAb Vaccination can play a positive role.
