目的探讨慢性粒细胞白血病(Chronic myelogenous leukemia,CML)来源的树突状细胞(Dendritic cell,DC)激活的T淋巴细胞对自体CML细胞的杀伤作用。方法利用rhGM-CSF、rhIL-4和rhTNF-α体外定向诱导CML患者外周血单核细胞(Peripheral blood mononuclear cell,PBMC)生成DC,光学显微镜和透射电镜观察所诱导的DC形态特征;流式细胞术进行表型检测;采用FISH和RT-PCR法检测所诱导的DC的白血病源性;MTT法检测其对自体CML细胞的杀伤作用。结果 CML患者PBMC在体外可诱导生成bcr/abl融合基因阳性的DC,其激活的T淋巴细胞对自体CML细胞的杀伤作用明显强于单独应用T淋巴细胞对CML细胞的杀伤作用。结论 CML患者PBMC能够诱导生成白血病源的DC,该DC能够激活T淋巴细胞对CML细胞的特异性杀伤作用。
Objective To investigate the cytotoxicity of T lymphocytes activated by dendritic cells (DCs) derived from chronic myelogenous leukemia (CML) on autologous CML cells. Methods Peripheral blood mononuclear cells (PBMCs) were induced by rhGM-CSF, rhIL-4 and rhTNF-α to induce DC in DCs. The morphology of DCs induced by rhGM-CSF, rhIL-4 and rhTNF-α were observed by light microscopy and transmission electron microscopy. Flow cytometry The phenotypes were detected. The induced leukemic origin of DCs was detected by FISH and RT-PCR. The cytotoxicity against autologous CML cells was detected by MTT assay. Results In vitro, PBMCs from patients with CML were induced to produce DC with positive bcr / abl fusion gene. The cytotoxicity of activated T lymphocytes to autologous CML cells was significantly stronger than that of T lymphocytes alone. Conclusion PBMCs from CML patients can induce the generation of leukemia-derived DCs, which can activate the specific killing effect of T lymphocytes on CML cells.
