目的探讨叶下珠复方II号(CPU II)对体外培养的人肝癌细胞HepG2增殖和凋亡的作用。方法采用四甲基偶氮唑盐比色法(MTT法),比较不同浓度的叶下珠复方在不同作用时间对体外培养的HepG2细胞增殖的影响;应用流式细胞术(FCM)结合Annexin V-FITC/PI荧光双染法观察CPU II诱导24h后人肝癌HepG2细胞的凋亡率。结果 MTT结果显示,人肝癌HepG2细胞经叶下珠复方II号处理一定时间后,增殖比率明显下降,2.60mg/ml以上的叶下珠II号对人肝癌HepG2细胞株的增殖有抑制作用(P<0.01;48h P<0.05),且呈一定的量效关系;FCM结果显示,随着叶下珠复方II号浓度的增加(2.6、26.0、130.0mg/ml),细胞的凋亡率也逐渐增加,差异有统计学意义(P<0.01)。结论叶下珠复方II号能够抑制人肝癌HepG2细胞生长和增殖,诱导人肝癌HepG2细胞凋亡,呈明显的量效关系。
Objective To investigate the effect of Ye Xia Zhu II on the proliferation and apoptosis of HepG2 cells cultured in vitro. Methods MTT method was used to compare the effects of different concentrations of Phyllanthus urinaria on the proliferation of HepG2 cells cultured in vitro. Flow cytometry (FCM) combined with Annexin V The apoptosis rate of human hepatocellular carcinoma HepG2 cells induced by CPU II was observed by FITC / PI double staining. Results The results of MTT assay showed that the proliferation rate of HepG2 cells was significantly decreased after treated with Phyllanthus Compound II for a certain period of time, and Yexiazhu II above 2.60mg / ml inhibited the proliferation of HepG2 cells (P <0.01; 48h, P <0.05), and showed a certain dose-effect relationship. The results of FCM showed that with the increase of Phyllanthus Compound II (2.6,26.0,130.0mg / ml) Increase, the difference was statistically significant (P <0.01). Conclusion Ye Xia Zhu Compound II can inhibit the growth and proliferation of human hepatoma HepG2 cells and induce the apoptosis of HepG2 cells in a dose-dependent manner.
