目的研究芦丁对雌性青春期大鼠内分泌激素水平、激素受体的表达以及乳腺组织形态学变化的影响,探讨芦丁促进乳腺发育的类雌激素样作用。方法 24只雌性青春期Wistar大鼠,随机分为3组,对照(Con):灌胃生理盐水2 ml/d;芦丁(Rut):灌服芦丁60 mg/(kg bw.d)一次;雌二醇(Est):肌注雌二醇60μg/(kg bw.w)。试验期2 w,结束时检测大鼠血浆和乳腺组织中雌激素(E2)、孕激素(P)、催乳素(PRL)、生长激素(GH)含量及其受体表达,并进行乳腺组织形态学观察。结果大鼠血浆与乳腺组织中E2、PRL水平Est组>Rut组>Con组(P<0.05);P水平Rut组>Con组(P<0.05),而与Est组差异不显著(P>0.05);GH水平Rut组与Con组与Est组差异不显著(P>0.05)。雌激素受体ER、孕激素受体PR、催乳素受体PRLR、生长激素受体GHR在乳腺组织中的表达量Rut组>Con组(P<0.05),Con组(P<0.05),与Est组差异不显著(P>0.05)。结论芦丁能够提高雌性青春期大鼠E2、P水平,促进PRL、GH分泌,上调乳腺组织中ER、PR、PRLR、GHR的表达量,促进乳腺发育。
Objective To study the effect of rutin on the level of endocrine hormone, the expression of hormone receptor and the morphological changes of mammary gland in female adolescent rats, and to explore the estrogen-like effect of rutin on mammary gland development. Conclusions Rutin can increase the levels of E2 and P in female adolescent rats, promote the secretion of PRL and GH, upregulate the expressions of ER, PR, PRLR and GHR in mammary gland tissues and promote the development of mammary gland.
