葡萄糖调节蛋白78(glucose-regulated protein 78,GRP78)是内质网应激(endoplasmic reticulum stress)相关的重要分子伴侣,涉及到多种肿瘤的增殖、存活、转移等演化过程。作为一种应激蛋白,GRP78在机体正常组织中几乎不表达,而在处于缺氧应激状态下的肿瘤组织中高表达,常常预示着不良的预后。GRP78还增强肿瘤对多种化疗的抵抗。因此GRP78的表达可作为预测肿瘤预后及化疗反应敏感度的标志物。此外,该蛋白质位于癌细胞表面的特性使它成为了一个很好的分子治疗靶点。
Glucose regulated protein 78 (GRP78) is an important molecular chaperone associated with endoplasmic reticulum stress, which involves the proliferation, survival and metastasis of many tumors. As a stress protein, GRP78 is almost not expressed in the normal tissues of the body, but highly expressed in tumor tissues under hypoxia stress, which often indicates poor prognosis. GRP78 also enhances tumor resistance to multiple chemotherapies. Therefore, GRP78 expression can be used as a predictor of tumor prognosis and chemosensitivity response markers. In addition, the properties of the protein on the surface of cancer cells make it a good molecular therapeutic target.
