目的:观察黄连解毒汤含药血清对脂多糖(LPS)诱导RAW264.7巨噬细胞炎症因子表达的影响。方法:以大、中、小剂量黄连解毒汤水煎剂灌胃SD大鼠制备含药血清,采用MTT法观察含药血清对细胞增殖的影响,选择各剂量浓度为20%的含药血清体外干预LPS刺激的巨噬细胞;采用Griess法检测NO释放量,采用ELISA法检测细胞上清液中TNF-α及IL-6含量。结果:采用LPS刺激细胞后,引起NO、TNF-α及IL-6因子的高表达,与正常对照组比较差异具有统计学意义(P<0.05);采用黄连解毒汤含药血清干预后,可明显抑制NO、TNF-α及IL-6因子的高表达,与模型组比较差异具有统计学意义(P<0.05或P<0.01)。结论:黄连解毒汤含药血清可抑制LPS诱导的巨噬细胞炎症反应,可能是该方防治动脉粥样硬化的重要机制。
Objective: To observe the effect of serum containing Huanglian Jiedu Decoction on lipopolysaccharide (LPS) -induced RAW264.7 macrophage inflammatory factor expression. Methods: SD rats were dosed with large, medium and small doses of Huanglian Jiedu Decoction to prepare drug-containing serum. MTT assay was used to observe the effect of drug-containing serum on cell proliferation. Drug-containing serum concentrations of 20% LPS-stimulated macrophages; NO release was detected by Griess assay, and the levels of TNF-α and IL-6 in supernatants were detected by ELISA. Results: After stimulated by LPS, the expression of NO, TNF-α and IL-6 were significantly higher than those in normal control group (P <0.05). After intervention with serum containing Huanglian Jiedu Decoction, Significantly inhibited the high expression of NO, TNF-α and IL-6, which had statistical significance compared with the model group (P <0.05 or P <0.01). Conclusion: Huanglian Jiedu Decoction serum can inhibit LPS-induced macrophage inflammatory reaction, which may be the important mechanism of the prevention and treatment of atherosclerosis.
