本文研究57例结肠癌患者体内产生前列腺素(PG)的抑制性单核细胞对植物血凝素(PHA)和刀豆素A(Con A)诱导的增殖反应,并用55名正常人作为对照。研究目的为确定结肠癌患者的T细胞对有丝分裂素的增殖反应是否降低,及该反应降低与PG介导的抑制作用是否有关。实验以患者外周血单核细胞(PBMC)为材料。将PBMC加入1μg/ml浓度的消炎痛或0.01%浓度的乙醇后放在微滴定平板上,再加入25μg/ml PHA或5μg/ml ConA孵育72小时。另外,用一定浓度的MCPB培养48小时后,离心取得上清液,用竞争性抑制放射免疫法测定其PGE_2含量。
This study investigated the proliferative responses induced by prostaglandin (PG)-inhibited mononuclear cells in 57 patients with colon cancer to phytohemagglutinin (PHA) and concanavalin A (Con A) and used 55 normal controls as controls. The purpose of the study was to determine whether the proliferative response to mitogen in T cells from patients with colon cancer was reduced and whether this response was associated with PG-mediated inhibition. The experiment used patient peripheral blood mononuclear cells (PBMC) as material. PBMCs were added to 1 μg/ml concentrations of indomethacin or 0.01% ethanol, placed on microtiter plates, and then incubated with 25 μg/ml PHA or 5 μg/ml ConA for 72 hours. In addition, after culturing with a certain concentration of MCPB for 48 hours, the supernatant was centrifuged and the PGE 2 content was measured by competitive inhibition radioimmunoassay.
