国外文献报道2,4-二氨基-5-取代苄基嘧啶类化合物(TMP类似物)具有一定的抑菌活性,为了研究这类化合物结构与活性的定量关系(QSAR),需要准确地测定它们的最低抑菌浓度(MIC),目前国内外沿用的测定方法有试管法和平皿法,但只测得一位有效数字,不能满足QSAR计算的要求,本文改进了MIC的经典测定法,用试管法培养,精确地配制药液浓度,采用分光光度法测定细菌浊度,计算得出三位有效数字的MIC值。本方法重复性好,数据可靠,操作简便,测得32个TMP类似物的MIC值,满足了定量构效关系的计算要求,为抑菌药物的定量构效关系研究开辟了新途径。
Foreign literature reports 2,4-diamino-5-substituted benzyl pyrimidine compounds (TMP analogs) have some antibacterial activity, in order to study the quantitative relationship between structure and activity of these compounds (QSAR), the need for accurate determination of them The minimum inhibitory concentration (MIC), the current domestic and international measurement methods are test tube and plate method, but only measured a valid number, can not meet the QSAR calculation requirements, this article improved the MIC classic determination method, with a test tube France culture, the precise preparation of liquid concentration, the use of spectrophotometric determination of bacterial turbidity, calculate the MIC of three significant figures. The method has good repeatability, reliable data and easy operation. The MIC values of 32 TMP analogues are measured, which meet the calculation requirements of quantitative structure-activity relationship and open up new ways for the quantitative structure-activity relationship study of antibacterial drugs.
