目的:研究慢性脑低灌注大鼠海马核因子E2相关因子2(Nrf2)的表达,以及当归注射液对其的作用。方法:双侧颈总动脉结扎(2VO)术式制备慢性脑低灌注大鼠模型共25只,随机分为生理盐水组(9只)、当归治疗组(9只)和假手术对照组(7只)。自2VO术后5周起,当归治疗组给予当归注射液腹腔注射,另两组给予生理盐水,共持续4周。之后取材,进行实时定量PCR与Westernblotting检测Nrf2的表达。结果:在2VO术后8周制备的大鼠慢性脑低灌注模型中,大鼠海马区Nrf2的RNA与蛋白水平表达与假手术对照组相比无显著差异(P>0.05)。当归治疗组大鼠海马的Nrf2RNA及蛋白水平表达均增高(P<0.05)。结论:在2VO术后8周,大鼠海马区Nrf2的RNA与蛋白水平表达改变不明显,当归注射液可以使Nrf2RNA与蛋白水平的表达均上调,可能是其改善慢性脑低灌注氧化性损伤的机制之一。
Objective: To study the expression of Nrf2 in the hippocampus of chronic cerebral hypoperfusion rats and the effect of angelica injection on it. Methods: Twenty-five rats were randomly divided into normal saline group (n = 9), Angelica sinensis group (n = 9) and sham operation control group (n = 7) only). Since 2VO 5 weeks after the operation, Angelica treatment group given intraperitoneal injection of Angelica injection, the other two groups were given saline for a total of 4 weeks. Afterwards, the expression of Nrf2 was detected by real-time quantitative PCR and Western blotting. Results: In chronic hypoxic-ischemic rat model induced by 2VO for 8 weeks, the expression of Nrf2 mRNA and protein in hippocampus of rats had no significant difference (P> 0.05) compared with the sham operation group. Angelica treatment group rats hippocampal Nrf2RNA and protein levels were increased (P <0.05). Conclusion: After 8 weeks of operation, the expression of Nrf2 mRNA and protein in hippocampus of rats did not change obviously. Angelica injection could up-regulate the expression of Nrf2 mRNA and protein, which may be related to the improvement of oxidative damage in chronic cerebral hypoperfusion One of the mechanisms.
