目的探讨不同浓度丁酸钠(NaB)对干扰素调节因子5(IRF5)基因表达的影响。方法体外培养人肺腺癌细胞A549和人胚肾细胞HEK293,分别用去离子水(对照组)和浓度为1.0、2.5和5.0mmol/L的NaB处理细胞(实验组),采用双荧光素酶报告基因技术检测IRF5基因启动子活性,采用RT-qPCR及Western blot法检测IRF5基因mRNA及蛋白表达水平。结果与对照组相比,实验组NaB可降低A549细胞及HEK293细胞中IRF5基因启动子活性、抑制A549细胞中IRF5基因mRNA及蛋白的表达(P<0.05或P<0.01)。随着NaB处理浓度的增加,IRF5基因启动子活性、mRNA及蛋白表达均呈下降趋势。结论 NaB对体外IRF5基因表达具有抑制作用。
Objective To investigate the effect of different concentrations of sodium butyrate (NaB) on the expression of interferon regulatory factor 5 (IRF5) gene. Methods Human lung adenocarcinoma cell line A549 and human embryonic kidney cell line HEK293 were cultured in vitro. The cells were treated with deionized water (control group) and NaB at concentrations of 1.0, 2.5 and 5.0 mmol / L, respectively. The cells were treated with dual luciferase The reporter gene was used to detect the promoter activity of IRF5 gene. The expression of IRF5 mRNA and protein was detected by RT-qPCR and Western blot. Results Compared with the control group, NaB in the experimental group decreased the IRF5 gene promoter activity and the mRNA and protein expression of IRF5 in A549 cells and HEK293 cells (P <0.05 or P <0.01). With the increase of NaB concentration, the promoter activity of IRF5 gene, mRNA and protein expression showed a downward trend. Conclusion NaB has an inhibitory effect on IRF5 gene expression in vitro.
