目的观察扇贝多肽(PCF)对紫外线A(UVA)照射后HaCaT角质形成细胞Fas、c-fos表达的影响,以探讨PCF保护UVA致HaCaT细胞损伤的分子机制。方法体外培养的HaCaT角质形成细胞分为5组:对照组、UVA模型组、5.69mmol/LPCF组、2.84mmol/LPCF组、1.42mmol/LPCF组。蛋白质印迹法检测Fas、c-fos蛋白表达;RT-PCR检测FasmRNA表达;荧光定量PCR检测c-fosmRNA表达。结果 UVA照射后模型组Fas、c-fos表达均显著增加(P<0.01),1.42~5.69mmol/L剂量范围内的PCF可抑制UVA引起的HaCaT细胞Fas、c-fosmRNA及蛋白表达(P<0.05),且有剂量依赖性。结论 PCF可通过抑制Fas、c-fos表达而抑制UVA诱导的HaCaT细胞损伤。
Objective To investigate the effect of scallop polypeptide (PCF) on the expression of Fas and c-fos in HaCaT keratinocytes after UVA irradiation, and to explore the molecular mechanism of PCF protecting HaCaT cells from damage induced by UVA. Methods HaCaT keratinocytes cultured in vitro were divided into 5 groups: control group, UVA model group, 5.69mmol / LPCF group, 2.84mmol / LPCF group and 1.42mmol / LPCF group. The expression of Fas and c-fos protein was detected by Western blotting. The expression of Fas mRNA was detected by RT-PCR and the expression of c-fos mRNA by fluorescence quantitative PCR. Results The expression of Fas and c-fos in HaCaT cells was significantly increased after UVA irradiation (P <0.01). PCF in the range of 1.42 ~ 5.69 mmol / L inhibited the expression of Fas and c-fos mRNA and protein in HaCaT cells induced by UVA (P < 0.05), and dose-dependent. Conclusion PCF can inhibit UVA-induced HaCaT cell injury by inhibiting the expression of Fas and c-fos.
