应用改性甲醇—水为流动相,在YwG—C_(18)H_(37)反相柱上,以普鲁卡因为内标,建立了适合临床血药浓度监测的高效液相色谱法。本法简便、快速,灵敏,重现性好。氯胺酮和普鲁卡因的平均回收率分别为97.97±3.7%和102.11±2.5%,不同浓度水平测定结果的日内和日间变异系数小于5%,其检定限度(信噪比>2)为0.3μg/ml。
The modified methanol - water was used as mobile phase. On the YwG-C 18 H 37 reverse phase column, procaine was used as internal standard to establish a HPLC method suitable for the monitoring of clinical plasma concentration. This method is simple, fast, sensitive and reproducible. The average recoveries of ketamine and procaine were 97.97 ± 3.7% and 102.11 ± 2.5%, respectively. The intra-and inter-day CVs of different concentration levels were less than 5% and the limits of detection (signal to noise ratio> 2) were 0.3 μg / ml.
