目的探讨来氟米特(leflunomide,LEF)对子宫内膜异位症大鼠模型中病灶体积及子宫内膜组织中转化生长因子β1(TGF-β1)的影响。方法采用自体移植法建立子宫内膜异位症大鼠模型。将SD大鼠分为对照组、生理盐水组(生理盐水1ml·kg-1·d-1灌胃)和LEF组(来氟米特35mg·kg-1·d-1灌胃),计算移植病灶体积,并采用免疫组化SP法检测异位病灶组织中TGF-β1的表达。结果 LEF组用药后异位病灶体积[(71.98±11.71)mm3]与对照组[(121.09±25.59)mm3]比较,差异有统计学意义(P<0.05);LEF组用药后异位病灶体积与生理盐水组[(90.19±10.0)mm3]比较,差异有统计学意义(P<0.05);LEF组用药后TGF-β1表达(103.41±3.31)与对照组(181.63±4.90)和生理盐水组(176.92±5.21)比较,差异有统计学意义(P<0.05)。结论 LEF可能通过抑制TGF-β1抑制子宫内膜异位病灶的生长,有望成为治疗子宫内膜异位症的新药物。
Objective To investigate the influence of leflunomide (LEF) on the volume of lesions and the expression of transforming growth factor-β1 (TGF-β1) in endometriotic rat model of endometriosis. Methods Autologous transplantation was used to establish a rat model of endometriosis. SD rats were divided into control group, saline group (1ml · kg-1 · d-1 normal saline) and LEF group (leflunomide 35mg · kg-1 · d-1) The volume of lesion was measured. The expression of TGF-β1 in ectopic lesions was detected by immunohistochemical SP method. Results Compared with the control group [(121.09 ± 25.59) mm3], the volume of ectopic lesion in LEF group was statistically significant (P <0.05). The volume of ectopic lesion in LEF group was significantly lower than that in control group [(71.98 ± 11.71) mm3] (90.19 ± 10.0) mm3], the difference was statistically significant (P <0.05). The expression of TGF-β1 in the LEF group was significantly higher than that in the control group (103.41 ± 3.31 vs 181.63 ± 4.90 vs the saline group 176.92 ± 5.21), the difference was statistically significant (P <0.05). Conclusion LEF may be a new drug for the treatment of endometriosis by inhibiting the growth of TGF-β1 inhibiting endometriosis lesions.
