纳米SiO_2和纳米TiO_2对雄性大鼠生殖毒性的实验研究

目的探讨纳米二氧化硅(SiO_2)和纳米二氧化钛(TiO_2)对雄性大鼠的生殖毒性。方法将56只SPF级SD雄性大鼠随机分为对照组、纳米SiO_2高、中、低剂量染毒组、纳米TiO_2高、中、低剂量染毒组。采用非暴露式气管滴注法染毒,高、中、低剂量染毒组染毒剂量分别为13.5 mg/kg、4.5 mg/kg、1.5 mg/kg,隔日染毒1次。4周后处死大鼠,HE染色法检测大鼠睾丸组织病理形态学改变,精子涂片法检测精子畸形、数量及存活率,ELISA法检测大鼠睾丸内睾酮含量。结果纳米SiO_2染毒组和纳米TiO_2染毒大鼠睾丸组织发生明显的病理学改变,且纳米TiO_2染毒组大鼠睾丸组织病理损伤明显强于纳米SiO_2染毒组。与对照组比较,纳米SiO_2高、中剂量染毒组及纳米TiO_2高、中剂量染毒组大鼠精子数量及存活率均降低,精子畸形率升高,差异均有统计学意义(P<0.05),且纳米TiO_2高剂量染毒组大鼠的精子数量、存活率均低于纳米SiO_2高剂量染毒组,纳米TiO_2高、中剂量染毒组大鼠的精子畸形率均高于纳米SiO_2高剂、中量染毒组,差异均有统计学意义(P<0.05)。纳米SiO_2高、中剂量染毒组和纳米TiO_2各染毒组大鼠睾酮的含量均低于对照组,差异有统计学意义(P<0.05)。结论纳米SiO_2和TiO_2均对雄性大鼠有生殖毒性效应,且纳米TiO_2的生殖毒性效应明显强于纳米SiO_2。 Objective To investigate the reproductive toxicity of nano-silica (SiO 2) and nano-TiO 2 in male rats. Methods Fifty-six SPF SD male rats were randomly divided into control group, high, medium and low dose Nano-SiO 2 exposure groups and high, medium and low dose Nano TiO 2 exposure groups. In non-exposed tracheal instillation, the doses of high, medium and low dose exposure groups were 13.5 mg / kg, 4.5 mg / kg and 1.5 mg / kg, respectively. After 4 weeks, the rats were sacrificed, the histopathological changes of testis were observed by HE staining, the sperm deformity, number and survival rate were detected by sperm smear method. The content of testosterone in testis of rats was detected by ELISA. Results The pathological changes were observed in the testis of rats exposed to nano-SiO 2 and nano-TiO 2. The pathological changes of testis in the nano-TiO 2 -treated group were significantly stronger than that of the nano-SiO 2 -treated group. Compared with the control group, the number and survival rate of sperm and the rate of sperm deformity in high and middle dose Nano-SiO 2 and medium-dose nano-TiO 2 groups were significantly decreased (P <0.05 ), And the number of sperm and survival rate of rats exposed to high dose of nano-TiO_2 were lower than that of high-dose nano-SiO_2 group. The sperm deformity rate of rats exposed to high and medium dose of nano-TiO_2 was higher than that of nano-SiO_2 Dose, the amount of exposure group, the difference was statistically significant (P <0.05). The contents of testosterone in the nano-SiO 2 high and medium dose groups and the nano-TiO 2 groups were lower than those in the control group (P <0.05). Conclusion Both nano-SiO_2 and TiO_2 have reproductive toxic effects on male rats, and the reproductive toxicity of nano-TiO_2 is obviously stronger than that of nano-SiO_2.
