目的:优化D-半乳糖致小鼠亚急性衰老实验中,腹腔注射法和皮下注射法的最佳造模剂量。方法:8周龄雄性ICR小鼠随机分为8组,分别为正常对照组、腹腔注射D-半乳糖组(1.25g/kg、2.5g/kg和5.0g/k)和腹腔注射D-半乳糖联合亚硝酸钠组(1.25g/kg+0.08g/kg-Na NO2),皮下注射组D-半乳糖组(0.125g/kg和0.5g/kg)和皮下注射组D-半乳糖联合亚硝酸钠组(0.125g/kg+0.08g/kg)。连续给药第7周测定各组小鼠空腹血糖情况,8周后解剖,计算脏器指数,检测血清、脑组织和肝脏组织中生化指标变化,同时对肝脏进行石蜡切片。结果:腹腔注射D-半乳糖2.5g/kg可显著提高血清中TC和TG含量,降低脑组织中GSH水平;皮下注射D-半乳糖0.5g/kg显著提高血清中TC、TG和脑组织中MDA含量,并降低脑中GSH水平;肝脏石蜡切片显示以上两组的小鼠肝组织排列疏松,体现衰老状态。结论:腹腔注射D-半乳糖2.5g/kg和皮下注射D-半乳糖0.5g/kg使小鼠整体表现出衰老症状,可分别作为D-半乳糖致小鼠亚急性衰老腹腔注射和皮下注射的最佳造模剂量。
Objective: To optimize the dosage of intraperitoneal injection and subcutaneous injection in D-galactose-induced subacute aging mice. METHODS: Male 8-week-old male ICR mice were randomly divided into 8 groups: normal control group, intraperitoneal injection of D-galactose (1.25g / kg, 2.5g / kg and 5.0g / Lactose combined with sodium nitrite group (1.25g / kg + 0.08g / kg-NaNO2), subcutaneous injection of D-galactose group (0.125g / kg and 0.5g / kg) Sodium nitrate group (0.125 g / kg + 0.08 g / kg). Results: Intraperitoneal injection of 2.5g / kg D-galactose significantly increased TC and TG levels in serum and GSH levels in brain tissue. Subcutaneous injection of 0.5g / kg D-galactose significantly increased serum TC, TG and brain tissue MDA content and reduce brain GSH levels; liver paraffin sections showed that mice in the above two groups of loosely arranged liver tissue, reflecting the state of aging. Conclusion: The intraperitoneal injection of 2.5g / kg of D-galactose and 0.5g / kg of D-galactose subcutaneously can induce the mice to develop senescence as a whole, which can be used as sub-acute intraperitoneal injection and subcutaneous injection of D-galactose The best modeling dose.
