目的:探讨核心蛋白聚糖Decorin对骨肉瘤细胞生长的影响及其作用机制。方法:用不同浓度的Decorin、TGF-β1作用于人成骨肉瘤细胞Saos-2,通过RT-PCR、Western Blot、MTT等方法检测其细胞增殖的情况及其凋亡的改变,并用流式细胞仪分析细胞的凋亡情况。结果:Decorin在体外能抑制骨肉瘤细胞的增殖,诱导骨肉瘤细胞凋亡,并且凋亡的出现是通过促进Fas/FasL来实现的。结论:Decorin可以通过促进Fas/FasL的表达促进细胞凋亡,进一步明确了Decorin作为抗肿瘤药物的可能。
Objective: To investigate the effect of decorin on the growth of osteosarcoma cells and its mechanism. Methods: The human osteosarcoma cell line Saos-2 was treated with different concentrations of Decorin and TGF-β1. The cell proliferation and apoptosis were detected by RT-PCR, Western Blot and MTT. Flow cytometry Analysis of cell apoptosis. Results: Decorin could inhibit the proliferation of osteosarcoma cells in vitro and induce the apoptosis of osteosarcoma cells, and the appearance of apoptosis was promoted by the promotion of Fas / FasL. Conclusion: Decorin can promote apoptosis by promoting the expression of Fas / FasL and further clarify the possibility of Decorin as antitumor drug.
