本研究用MDR_1基因为探针,用Northern blot的方法观察了肝癌组织癌旁肝组织及化疗前后肝癌组织之间MDR_1基因表达的差别。结果发现:肝癌组织中MDR_1基因的表达高于癌旁肝组织;化疗后肝癌组织中MDR_1基因的表达也高于未化疗肝癌组织。提示MDR_1基因的过量表达可能与肝癌固有的和获得性抗药性密切相关。本文还报道了用多聚酶链反应定量分析肝癌组织中MDR_1基因表达的方法。
In this study, the MDR-1 gene was used as a probe, and Northern blot was used to observe the difference of MDR-1 gene expression between adjacent hepatocellular carcinoma tissue and before and after chemotherapy. The results showed that the expression of MDR1 gene in hepatocellular carcinoma was higher than that in paracancerous liver tissues. The expression of MDR1 gene in hepatocellular carcinoma tissues after chemotherapy was also higher than that in non-chemotherapy liver cancer tissues. It suggests that the overexpression of MDR_1 gene may be closely related to the inherent and acquired drug resistance of liver cancer. This paper also reported a method for quantitative analysis of MDR1 gene expression in hepatocellular carcinoma using polymerase chain reaction.
