目的:探讨NDRG2蛋白在非小细胞肺癌(NSCLC)组织中的表达及其在病理分类、分级、TNM分期中的差异性,为进一步研究NDRG2在NSCLC中的作用及其机制提供依据。方法:采用免疫组化EnVinsion法检测NDRG2蛋白在155例非小细胞肺癌中的表达,统计学分析其病理分类、分级及临床TNM分期的差异性。结果:NDRG2阳性表达主要定位于胞质中和胞膜上,在癌和相应正常肺组织中总阳性率分别为63.23%、100%,两者相比差异性非常显著(P=0.0000),其中肺鳞癌和腺癌阳性率分别为54.65%、73.91%,与正常组织比差异性均非常显著(P=0.0000);鳞癌与腺癌间的阳性率和阳性强度差异性显著(P=0.0131),鳞癌高、中、低分化阳性率分别为87.10%、41.3%和11.11%,经Kruskal-Wallis H检验差异性显著(P=0.0000),腺癌的高、中、低分化的阳性率分别是100.0%、88.89%和31.58%,经检验差异性非常显著(P=0.0000);鳞癌、腺癌的TNM分期Ⅰ、Ⅱa、Ⅱb、Ⅲa、Ⅲb的阳性率有逐渐降低的趋势,经检验其差异性显著,P值分别为0.0005、0.0342。结论:NDRG2在NSCLC中低于相应正常组织中的表达,表达的高低与病理分级高低成正比、与TNM分期高低成反比,很可能对NSCLC发生、发展产生重要影响。
OBJECTIVE: To investigate the expression of NDRG2 in non-small cell lung cancer (NSCLC) and its difference in pathological classification, grading and TNM staging, so as to provide a basis for further study on the role and mechanism of NDRG2 in NSCLC. Methods: The expression of NDRG2 protein in 155 cases of non-small cell lung cancer was detected by immunohistochemical EnVionion method. The difference of pathological classification, grading and clinical TNM staging were statistically analyzed. Results: The positive expression of NDRG2 mainly localized in cytoplasm and membrane, and the positive rates of NDRG2 in cancer and corresponding normal lung tissues were 63.23% and 100%, respectively, with significant difference (P = 0.0000), among which The positive rates of squamous cell carcinoma and adenocarcinoma were 54.65% and 73.91%, respectively, which were significantly different from those of normal tissues (P = 0.0000). There was significant difference between positive rate and positive rate of squamous cell carcinoma and adenocarcinoma (P = 0.0131 ). The positive rates of high, moderate, and poorly differentiated squamous cell carcinoma were 87.10%, 41.3% and 11.11%, respectively. The positive rates of high, moderate and poorly differentiated adenocarcinoma were significantly different by Kruskal-Wallis H test (P = 0.0000) (P = 0.0000). The positive rates of TNM staging Ⅰ, Ⅱa, Ⅱb, Ⅲa and Ⅲb in squamous cell carcinoma and adenocarcinoma tended to decrease gradually Test significant differences, P values were 0.0005,0.0342. Conclusion: The expression of NDRG2 in NSCLC is lower than that in corresponding normal tissues. The expression of NDRG2 is directly proportional to the level of pathological grade. It is inversely proportional to the TNM stage, which may have a significant impact on the occurrence and development of NSCLC.
