观察二甲双胍对STZ糖尿病大鼠血糖和骨骼肌中葡萄糖转运体Ⅳ(GLUT4)基因表达的影响。方法STZ糖尿病大鼠每公斤体重100mg/d二甲双胍灌胃治疗3周后,斑点杂交测定骨骼肌中的GLUT4mRNA含量。结果糖尿病治疗组的血糖降低30.3%,较非治疗组低28.6%,而骨骼肌中GLUT4mRNA含量较非治疗组高65.6%。结论二甲双胍促进骨骼肌中GLUT4基因表达可能是其降低STZ糖尿病大鼠血糖的机制之一。
To observe the effect of metformin on blood glucose and glucose transporter Ⅳ (GLUT4) gene expression in STZ diabetic rats. Methods STZ diabetic rats were treated with 100 mg / d of metformin per kg body weight for 3 weeks. The content of GLUT4 mRNA in skeletal muscle was determined by dot blot hybridization. Results The diabetes group had a 30.3% decrease in blood glucose, 28.6% lower than the non-treatment group, and 65.6% higher in the skeletal muscle than in the untreated group. Conclusion Metformin promotes the expression of GLUT4 in skeletal muscle may be one of the mechanisms of lowering blood glucose in STZ diabetic rats.
