目的:通过观察萸连巴布剂的镇痛抗炎作用、刺激性及过敏性,对萸连巴布剂的药效学及安全性进行评价,为萸连巴布剂的临床应用提供实验依据。方法:角叉菜胶致炎法致大鼠足肿胀;化学刺激法造小鼠镇痛抗炎模型;分别观察外用萸连巴布剂高、中、低3个剂量(含生药12,6,3 g.kg-1外用涂抹)组对足跖肿胀度、扭体次数及毛细血管通透性的影响。健康家兔分别单次和多次给药,进行完整与破损皮肤的皮肤刺激性试验;健康豚鼠,通过致敏接触和激发接触给药进行皮肤过敏试验。结果:萸连巴布剂中、低剂量组能明显降低大鼠足肿胀度,中剂量组作用时间持续时间较长。高、中剂量组降低毛细血管通透性,高、中、低3个剂量组能减少扭体次数,与赋形剂组比较均有显著性差异(P<0.05,P<0.01)。单次给药和连续给药,完整皮肤和破损皮肤组均未出现红斑、水肿等刺激反应。豚鼠皮肤未出现过敏反应。结论:萸连巴布剂具有镇痛抗炎和消肿的作用,对家兔完整皮肤、破损皮肤均无刺激性,对豚鼠皮肤不会引起过敏反应。
OBJECTIVE: To observe the analgesic and anti-inflammatory effects, irritation and allergy of cornelian poultice to evaluate the pharmacodynamics and safety of cornelian poultice, providing the experimental basis for the clinical application of cornelian poultice . Methods: Carrageenan induced inflammation of rat foot caused by swelling; chemical stimulation of mouse analgesic model of anti-inflammatory; observed with topical buspian topical, high and low doses (including crude drug 12,6, 3 g.kg-1 topical application group on the foot swelling, writhing times and capillary permeability. Healthy rabbits were single and multiple administration, respectively, to complete and damaged skin irritation test; healthy guinea pigs, sensitized and challenged contact dermal allergy test. Results: Dogwood poultice, low-dose group can significantly reduce the degree of paw swelling in rats, the role of dose group longer duration. The high and middle dose groups decreased capillary permeability. The high, medium and low dose groups could reduce the number of writhing, which were significantly different from the vehicle group (P <0.05, P <0.01). A single administration and continuous administration, intact skin and damaged skin group did not appear erythema, edema and other stimuli. Guinea pig skin does not appear allergic reactions. CONCLUSION: The dogwood Lianbubu has analgesic, anti-inflammatory and anti-swelling effects. It has no irritation to intact skin and damaged skin of rabbits and does not cause allergic reaction to the guinea pig skin.
