目的 :探讨癌性胸水肿瘤浸润淋巴细胞 (TIL)的生物学特性 ,以便更好地应用于临床。方法 :用低浓度白细胞介素 2 (IL - 2 )诱导培养癌性胸水TIL ,按常规法计数TIL细胞增殖量 ,采用流式细胞仪分析TIL细胞表型及MTT法检测其杀伤细胞活性。结果 :经低浓度IL - 2 (10 0IU/ml)诱导培养TIL 2 1d后 :TIL增殖倍数为2 130± 1140 ,大于 10 0 0倍占 72 %。CD3 无显著性变化 (P >0 .0 5 ) ,CD4 、CD8有显著性差异 (P <0 .0 1) ,而CD4 /CD8比值高达 3.5 3± 0 .82 ,并且在 2 1d时TIL具有较高的细胞毒性。结论 :癌性胸水TIL体外诱导培养3周时TIL具有较强的生物活性 ,此时进行胸水治疗可取得可靠的疗效。
Objective: To investigate the biological characteristics of cancerous pleural effusion tumor infiltrating lymphocytes (TIL) for better clinical application. METHODS: The pleural effusion TIL was cultured with low concentration of interleukin 2 (IL-2). The proliferation of TIL cells was counted according to conventional methods. Flow cytometry was used to analyze the phenotype of TIL cells and MTT assay was used to detect the activity of killer cells. RESULTS : After 1 day of TIL 2 induction induced by low concentration of IL - 2 (10 0IU/ml) : the proliferation of TIL was 2 130 ± 1140 and it was greater than 100 % of 72 % . There was no significant change in CD3 (P > 0.05), there was a significant difference between CD4 and CD8 (P < 0.01), while the CD4/CD8 ratio was as high as 3.53 ± 0.82, and TIL had a High cytotoxicity. Conclusion :TIL has a strong biological activity when cancer pleural effusion TIL is cultured in vitro for 3 weeks. At this time, pleural effusion therapy can achieve a reliable therapeutic effect.
