目的:探讨肿瘤坏死因子α(TNFα)对血管内皮细胞(VEC)损伤的机制以及在急性肺损伤(ALI)发病中的作用。方法:通过建立人脐静脉内皮细胞(HUVEC)体外培养,采用流式细胞技术观察TNFα对HUVEC凋亡的影响以及抗TNFα的保护效应。结果:TNFα加入细胞培养中,可明显诱导HUVEC凋亡,并随着作用时间的延长和TNFα浓度增加,细胞凋亡率也相应增加,一定范围内呈时间、剂量、效应关系。而抗TNFα单克隆抗体可明显阻断由TNFα诱导的细胞凋亡。结论:细胞凋亡是VEC受损的主要方式之一。
Objective: To investigate the mechanism of tumor necrosis factor α (TNFα) injury to vascular endothelial cells (VECs) and its role in the pathogenesis of acute lung injury (ALI). Methods: The human umbilical vein endothelial cells (HUVECs) were cultured in vitro. The effects of TNFα on the apoptosis of HUVECs and the protective effect of anti-TNFα were observed by flow cytometry. Results: The apoptosis of HUVEC induced by TNFα was obviously induced by TNFα. The apoptosis rate of HUVEC induced by HUVEC obviously increased with the prolongation of action time and the increase of TNFα concentration, and showed a time, dose and effect relationship within a certain range. Anti-TNFα monoclonal antibody can obviously block the apoptosis induced by TNFα. Conclusion: Apoptosis is one of the main ways of VEC damage.
