目的:通过观察摘除卵巢复制骨质疏松症模型大鼠股骨中BMP-2蛋白表达变化,探究补肾活血方能有效治疗质疏松症的机理。方法:实验采用摘除大鼠双侧卵巢的方法复制骨质疏松症模型,并将大鼠分为正常组、假手术组、模型组、补肾活血组、阳性药物组福善美组和骨疏康组,术后2 d开始给药干预,6次/周,持续12周后检测各组大鼠股骨中BMP-2蛋白表达水平及骨密度。结果:结果补肾活血组和骨疏康组、福善美组与模型组相比较,股骨密度都得到了一定提高(P<0.01);BMP-2蛋白表达水平也都有显著升高(P<0.01)。结论:补肾活血方对骨质疏松症模型大鼠有明显的治疗作用,其作用机制可能是在一定程度上提高了股骨组织中BMP-2的水平,进而抑制骨组织的吸收,促进骨组织的形成。
OBJECTIVE: To observe the changes of BMP-2 protein expression in the femur of ovariectomized osteoporosis model rats and explore the mechanism of Bushen Huoxue decoction in the treatment of osteoporosis. Methods: Osteoporosis model was duplicated by ovariectomized rats. The rats were divided into normal group, sham operation group, model group, Bushen Huoxue group, Fushanmei group and Gushukang group The intervention was started 2 days after the operation. Six times a week for 12 weeks, the BMP-2 protein expression and BMD in the femur of each group were detected. Results: Compared with the model group, the femoral bone density of Bushen Huoxue group, Gushukang group and FuShanMei group were all increased (P <0.01), and the expression of BMP-2 protein also increased significantly (P <0.01) ). Conclusion: Bushen Huoxue Recipe has a significant therapeutic effect on rats with osteoporosis, and its mechanism may be to a certain extent, increase the level of BMP-2 in femur tissue, and then inhibit the absorption of bone tissue and promote bone tissue form.
