目的观察神经病理痛大鼠背根神经节细胞钙通道α1B亚单位mRNA的表达。方法SD大鼠24只,随机分为3组,神经病理痛组:行左侧L5脊神经结扎并切断建立大鼠神经病理痛模型;假手术组:暴露左侧L5脊神经,但不行结扎和切断;正常对照组:不作手术处理,每组8只。取三组大鼠L5背根神经节,用原位杂交方法测定钙通道α1B亚单位mRNA表达。结果神经病理痛组背根神经节细胞钙通道α1B亚单位mRNA吸光度值为0.033±0.011,明显低于假手术组(0.065±0.042)和正常对照组(0.066±0.034)(P<0.05)。结论神经病理痛大鼠L5背根神经节细胞钙通道α1B亚单位mRNA表达水平的下降可能是背根神经节细胞钙电流降低及兴奋性增强的原因。
Objective To observe the mRNA expression of α1B subunit of calcium channel in dorsal root ganglion neurons in rats with neuropathic pain. Methods Twenty-four Sprague-Dawley rats were randomly divided into three groups. Neuropathic pain group: L5 spinal nerve was ligated and cut off to establish the model of neuropathic pain in rats. Sham-operated group: L5 spinal nerve was exposed on the left side but not ligated and severed; Normal control group: no surgery, 8 in each group. Three groups of L5 dorsal root ganglion were taken and the expression of calcium channel α1B subunit mRNA was measured by in situ hybridization. Results The mRNA absorbance of α1B subunit of Ca2 + channel in dorsal root ganglion neurons was 0.033 ± 0.011 in neuropathic pain group, which was significantly lower than that in sham operation group (0.065 ± 0.042) and normal control group (0.066 ± 0.034) (P <0.05). Conclusions The decrease of calcium channel α1B subunit mRNA expression in L5 rat dorsal root ganglion neuropathic pain rats may be the reason of the decrease of calcium current and excitability of dorsal root ganglion cells.
