目的:观察柿叶总黄酮(PLF)对大鼠心肌缺血再灌注(MIR)损伤的保护作用。方法:采用在体大鼠结扎冠状动脉左前降支30min后再灌注60min造成心肌缺血再灌注模型。在结扎前30min分别用不同剂量PLF进行预处理。测定血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、天冬氨酸转氨酶(AST)、乳酸脱氢酶(LDH)、乳酸脱氢酶同工酶(LDH1)、肌酸磷酸激酶(CK)和肌酸磷酸激酶同工酶(CK-MB)的变化,测定心肌组织中ATP酶的活力。结果:与MIR组相比,PLF预处理能明显降低血清中MDA、AST、LDH、LDH1、CK和CK-MB的含量,提高SOD、GSH-Px的活性,同时提高心肌组织中ATP酶活性。结论:PLF对大鼠MIR损伤具有显著的保护作用,其机制可能与抗自由基,保护氧自由基清除酶活性和改善缺血心肌能量代谢障碍有关。
Objective: To observe the protective effect of persimmon leaf flavonoids (PLF) on myocardial ischemia-reperfusion (MIR) injury in rats. Methods: Myocardial ischemia / reperfusion model was induced by ligating the left anterior descending artery of the coronary artery for 30 min in rats and then reperfusion for 60 min. 30min before ligation with different doses of PLF pretreatment. Serum levels of MDA, SOD, GSH-Px, AST, LDH, LDH1, CK and CK-MB were measured to determine the activity of ATPase in myocardium. Results: Compared with MIR group, PLF preconditioning could significantly decrease the levels of MDA, AST, LDH, LDH1, CK and CK-MB in serum, increase the activity of SOD and GSH-Px, and increase the activity of ATPase in myocardium. CONCLUSION: PLF has a significant protective effect on MIR injury in rats. The mechanism may be related to anti-free radical, protecting free radical scavenging enzyme activity and improving energy metabolism of ischemic myocardium.
